An Improved Method for Efficient DNA Extraction from Grapevine

Grapevine (Vitis vinifera L.) is one of the oldest and most important perennial crops worldwide which has been subject extensive genetic studies including gene mapping, transformation, DNA fingerprinting. Grapevines are rich in polysaccharides, polyphenolic compounds, various secondary metabolites, many have significant importance food, agrochemical, pharmaceutical industries. While metabolites indicators quality grapevines, presence them makes grapevine difficult plants to extract from. These not only affect extraction procedures but also downstream reactions such as restriction digestion PCR. Development new genotyping techniques based on sequencing by (GBS) requires high-quality for sequencing. To date, several protocols developed from grapevine. In this study, three different with modifications were compared performance leaves four cultivars. Efficiencies these methods determined extracted DNA’s quantity quality. confirm suitability GBS, was digested enzymes. Although all traditional CTAB method, they resulted yield efficiency.
